In evaluating drugs as candidates for sustained release formulation,the disadvantages of such formulation that must be considered includes the following
1.Administration of sustained release formulation that not permit prompt termination of therapy: Immediate changes in drug need during therapy, such as might be encountered if significant adverse effects are noted, cannot be accommodated.
2.Sustained released forms are designed for normal populations, I.e.,on the basis of average biologic half-lives, consequently, disease states that alters drug disposition, significant patient variation and so forth cannot be accommodated.
3. Economic factors must also be assessed, since more costly process and equipments are involved in manufacturing many sustained released forms
